Create a shortcut like this:
Target: %windir%\System32\rundll32.exe user32.dll,LockWorkStation
Start in: %windir%
Then, you can just click it to lock your desktop. It is really fast - even faster than quick user switching. Thus, this is excellent if you share a computer with someone else.

The NT line allows you to lock your workstation... the way I've always done it in the past with WinNT4 and Win2k is to hit ctrl+alt+del and hit the button "Lock Workstation" What this does is locks the workstation so that no one can come, sit down at it and have access to your files etc. You have to input a password to unlock the workstation. This is just a nice lil shortcut to let it happen quicker... altho I've gotten pretty quick at hitting ctrl+alt+del and then Enter really fast.
